特异引物PCR快速检测可疑牙周致病菌的相关性研究
Study on the specific-primer PCR detection of 3 species of putative periodontal pathogens and the correlation between the presence of bacteria and periodontosis

Abstract
Objective To establish a rapid multiple specific-primer PCR for detection of three species of bacteria Aa, Pi and Pg,and the correlation between the presence of bacteria and the periodontosis.Methods Subgingival microftora samples were collected from 30 patients with moderate to sever periodotitis and 30 periodontally healthy patients,the PCR was used to amplify the target genes.Results Sensitivity and specificity of PCR were also evaluated with the reference bacteria.The multiple PCR was able to detect a minimum of 3.25ng bacteria DNA.With good specificity,target fragments of 3 species of bacteria were detected.The prevalence of Pg in adults periodentosis were 86.7％ vs 13.3％ in the healthy group (P＜0.001).The prevalence of Pi in adults periodentosis were 70％ vs 20％ in the healthy group (P＜ 0.001 ),The prevalence of Aa in adults periodentosis were 6.7％ vs 3.3％ in the healthy group (P＞0.5).Conclusion As the putative periodontal pathogens of adult periodontitis,Pg,Pi,Aa may be part of the normal oral flora,that need futher study.关键词
