Expression of PCNA in pulmonary artery smooth muscle cells of hypoxic pulmonary hypertensive mice

Abstract
Objective To establish a mouse model of hypoxic pulmonary hypertension and to explore the expression of proliferating cell nuclear antigen (PCNA) in aorta and pulmonary artery smooth muscle cells of rats with hypoxic pulmonary hypertension.Methods SPF SD mice were randomly divided into 2 groups: normal group (n = 10) and hypoxia group (n = 10).The mouse model of hypoxic pulmonary hypertension was constructed by intermittent ordinary pressure hypoxia.Histology of HE stained lung tissue sections and image analysis were performed to detect the morphological changes of pulmonary arterioles.PCNA level in pulmonary artery smooth muscle cells were detected by immunohistochemistry,and the PCNA concentration was determined by semi-quantitative image analysis.Results Four weeks later, MT% and MA% in the SD mice of hypoxia group were significantly higher than that of normal group (P < 0.05).PCNA ribonucleoprotein expression ( area IOD) of pulmonary artery smooth muscle cells in the SD mice of hypoxia group was significantly higher than that of the normal group (P < 0.05).Conclusions The method of intermittent ordinary pressure hypoxia can be successfully used to construct a hypoxic pulmonary hypertensive mouse model within four weeks, suggesting that PCNA expression may play a very important role in the formation of chronic HPH.关键词
