Effects of electroacupuncture on levels of SOD and MDA in the brain of rats with cerebral ischemia reperfusion injury under different anesthesia

Abstract
Objective To observe the changes of superoxide dismutase (SOD), malonyldialdehyde(MDA) and PPAR - γmRNA expression in the brain tissues of rats with cerebral ischemia- reperfusion injury and investigate the effects of electroacupuncture on the levels of SOD,MDA and PPAR - γmRNA expression under different anesthesia. Methods Forty male SD rats were uniformly randomized into A, B, C, D and E groups with eight rats in each group. Transient general brain ischemia was induced by four - vessel occlusion and reperfusion under intravenous chloral hydrate or propofol anesthesia. EA (30 to 50 Hz, 1 mA) was applied to Baihui(GV20), Mingmen(GV4) and Zusanli(ST36) for 20 min after IR. The content of SOD and MDA and PPAR -γmRNA expression in the brain tissues were measured postischemia after 24 h. Results SOD levels in group A was significantly lower, while MDA levels and PPAR - γmRNA expression was significantly higher than E group (P<0.05, P<0.01, respectively); SOD levels in B and D group were found to have increased, while MDA levels and PPAR - γmRNA expression in groups C and D were found to have decreased in comparison with group E (P<0.05, P <0.01, respectively);SOD levels in group D were found to have increased in comparison with group C ( P < 0.05 ) ;MDA levels and PPAR - γmRNA expression in groups C and D were found to have decreased in comparison with group B (P <0.01). MDA was positive correlation with the expression of PPAR- γmRNA . And the correlation coefficient was 0. 664 ( P < 0.01 ). Conclusions Electroacupuncture combined with propofol can protect the brain of cerebral ischemia- repeffusion rat by electroacupuncture improving the activity of SOD and propofol decreasing MDA content and PPAR- γmRNA expression in the brain, and there was an inherent dependability between the MDA and PPAR - γmRNA expression.关键词
