Abstract
Purpose To investigate the specificity of monoclonal anti-p2lras antihody KGH-R1 in hreast cancer tissues. Methods Using monoclonal anti-p2lras antibody KGH-R1 prepared in our laboratory,immunohistochemical SP technique was performed in formalin-fixed, paraffin-embedded breast samples including 56 cases of normal breast tissues, 55 breast hyperplasia without atypia, 33 atypi cal breast ductal hyperplasia, 35 ductal carcmomas in situ and 60 invasive ductal carcinomas. Immunoreactivity of monoclonal antibody KGH-R1 was evaluated by percentage and histological score ( HSCORE ). Results Immunostaining was found in 71. 67％ ( 43/60 ) of invasive breast carcinomas with an average of 66. 29％ of carcinoma cells positive and average of 131. 64 of HSCOREs. There were not significant correlations between KGH-R1 immunoreactivity( HSCOREs ) and age, lymph node metastasis. ER or PR status( P ＞ 0. 05 ).62. 86％ ( 22/35 ) of ductal carcinoma in situ demonstrated immunostaining with KGH-R1 ; the average percentage of carcinoma cells positive was 59. 18％ ; the average HSCOREs was 86. 75. 57. 58％ ( 19/33 ) of atypical breast hyperplasia deruonstrate.d immunoreactivity with KGH-R1 ; the average percentage of atypical ductal hyperplasic cells positive was 46. 83％ ,and the average HSCOREs was 54. 86. 49. 09％( 27/55 ) of breast hyperplasia without atypia showed immunoreactivity with KGH-RI , the average percentage of hyperplasic ductal cells positive was 23. 15％ , and the average HSCOREs was 21. 95. In normal breast tissues, onlyfew( 12/56 ) samplesshowed weak immunoreactivity in ductal epithelial cells,and the HSCOREs of all positive samples were less than 30. Conclusions It is suggested in this study that monoclonal anti-p21 ras antibody KGH-R1 has a good specificity for breast cancers.关键词
