Experimental Study of Protective Effect of Intranasal Immunization with ΔA146 Ply Against Colonization of Streptococcus Pneumonia

Abstract
Objective This study was aimed to obtain △A146 Ply protein by prokaryotic expression and evaluate intranasal immunization with △A146 Ply against nasopharyngeal colonization of Streptococcus pneumonia ( S. pn ) . Methods △A146 Ply protein was obtained by induction with IPTG, and purification with Ni-NTA resin. BALB/C mice were intranasally immunized to obtain △A146 Ply specific antiserum. The in vivo anti-colonization of△A146 Ply protein against S. pn serotype 19F was evaluated by the number of surviving bacteria existing in nasopharyngeal washes and lung homogenates. In addition. multiple strains of serotype 14. 3, 6, and 2 were cultured to further determine the protective effect of△A146 Ply by in vitro anti-adhesion assay in A549 cells. Results △A146 Ply protein was obtained with >90% purity. In vivo experiments showed that intranasal immunization with △A146 Ply significantly reduced the colonization number of 19F Streptococcus pneumoniae in nasopharynx and lung of hosts ( P <0. 01 ) . Colonization of multiple serotypes 14 and 3 in the nasopharynx and lung of immunized mice was significantly decreased compared to control mice ( P < 0. 05 ) . Bacteria of serotype 2 colonized in the lung were significantly decreased ( P <0. 05 ) . Serotype 6B did not show difference in colonization in both immunized and control mice ( P >0. 05 ) . In vitro anti-adhesion results showed that the inhibition effect of △A146 Ply-specific antibody was dose-dependent. Conclusion Intranasal immunization of BALB/C mice with △A146 Ply protein could protect hosts against colonization of various Streptococcus pneumoniae serotypes in the nasopharynx as well as lung , suggesting Ply protein as pneumococcal vaccine candidate.关键词
