RP-HPLC法测定小鼠血浆中10-羟基喜树碱的浓度
Determination of the Plasma Concentration of 10-hydroxycamptothecin in Mice by RP-HPLC

Abstract
OBJECTIVE: To establish the method for the concentration determination of 10-hydroxycamptothecin (HCPT) in mice plasma.METHODS: 33 mice were given HCPT 80 mg· kg-1 intragastrically.The blood samples of heart were collected before administration and 0.17 h, 0.33 h, 0.67 h, 1 h, 1.5 h, 2 h, 4 h, 8 h, 12 h, 24 h after administration.The plasma concentration of HCPT was determined on Luna C18 column with mobile phase consisted of water-methanol (gradient elution) at the flow rate of 1.2 mL· min-1.Fluorescence detector with excitation and emission wavelengths of 383 nm and 553 nm was used for the determination of HCPT Camptothecin was used as internal standard.RESULTS: Endogenous impurity did not interfere with the determination.The linear range of HCPT were 1.25～80 ng·mL-1 (r=0.999 7).The average assay recovery was 96.46％～101.87％ and extraction recovery was 82.4％ ～109.6％.The intra-day and inter-day precision were less than 5.00％.CONCLUSION: The method is simple, reliable, specific and sensitive.It is suitable for plasma concentration of HCPT.关键词
