血管紧张素-(1-7)与血管紧张素Ⅱ对THP-1巨噬细胞NPC1表达及胆固醇流出的影响
Influence of angiotensin-(1-7) and angiotensin II on expression of NPC1 and cholesterol efflux in THP-1 macrophages

Abstract
AIM : To investigate the effects of angiotensin - ( 1 - 7 ) [ Ang - ( 1 - 7 ) ] and angiotensin Ⅱ ( Ang Ⅱ ) on the expression of NPCI in THP - 1 macrophages.The rate of cholesterol effluent was also observed.METHODS :The macrophages were derived from THP - 1 monocytes induced by phorbol myristate acetate for 48 h and then the cells were randomly divided into control group, Ang Ⅱ group , Ang - ( 1 - 7 ) group , Ang - ( 1 - 7 ) + Ang Ⅱ group , and Ang ( 1 -7 ) + Ang Ⅱ +A -779 [ a specific antagonist of Ang -( 1 -7 )receptor ] group.The expression of NPC1 at mRNA and protein levels was determined by reverse transcription - polymerase chain reaction ( RT - PCR ) and Western blotting,respectively.Cholesterol effluent was measured by liquid scintillation counting.RESULTS : Compared with control group,the expression of NPC1 at mRNA and protein levels was significantly down - regulated in Ang Ⅱ group and the effluent rate of cholesterol was also decreased ( P ＜0.05 ).Ang -( 1 -7 ) dose - dependently inhibited the reduction of NPC1 induced by Ang Ⅱ, and promoted the cholesterol efflux ( P ＜0.05 ).However, when incubated with A - 799 , the effect of Ang ( 1 - 7 ) on inhibiting the mRNA expression of NPC1 was significantly attenuated.CONCLUSION : The present study indicates that Ang -( 1 -7 ) down - regulates the expression of NPC1 dose - dependently in THP - 1 macrophages induced by Ang Ⅱ via its specific receptor Mas and contributes to the process of anti - atherosclerosis.关键词
