柴胡皂甙d对人肝癌细胞HIF-1α/COX-2信号通路的调节作用
The role of saikosaponin d in regulating HIF-1α/COX-2 signal transduction pathway in human hepatocellular carcinoma cells

Abstract
Objective To comparatively observe changes in the expressions of hypoxia inducible factor-1α (HIF-1α) and cyclooxygenase-2 (COX-2) in human hepatocellular carcinoma cells under hypoxic conditions as well as the interfering effect of saikosaponin d, and explore the influence of saikosaponin d on HIF-1α/COX-2 pathway.Methods ① Cobalt chloride was used to simulate cell hypoxia to observe the expressions of HIF-1α and COX-2 in human hepatocellular carcinoma cell line SMMC-7721 under hypoxic conditions. ② Cobalt chloride was used to simulate cell hypoxia. Rapamycin, the selective inhibitor of mTOR, was used to block the synthesis of HIF-1α protein. Then the changes of COX-2 expression in SMMC-7721 cell were observed. ③ Saikosaponin d of different concentrations were used to act on the hepatocellular carcinoma cell SMMC-7721 cultured under hypoxic conditions simulated by cobalt chloride. The expressions of HIF-1α and COX-2 were detected after 24 hours. Results ① In the normal control hepatocellular carci(\n)oma cell SMMC-7721, only a small amount of HIF-1α protein could be detected. Compared with HIF-1α, a certain amount of COX-2 protein could be detected in the same normal control group. Once exposed to hypoxic interference, the level of HIF-1α protein increased apparently. At the same time,the level of COX-2 protein increased after exposure to hypoxia. ② When SMMC-7721 cells cultured under hypoxic conditions were exposed to rapamycin, the synthesis of HIF-1α protein in the corresponding treatment groups was inhibited apparently. Meanwhile, the level of COX-2 protein decreased significantly. ③ Saikosaponin d inhibited the expression of HIF-1α protein in a dose-dependent manner under the hypoxic condition simulated by cobalt chloride. The protein level of COX-2 also declined with the increase in the concentration of saikosaponin d. Conclusion HIF-1α is involved in promoting the expression of COX-2 in hepatocellular carcinoma cells under hypoxic conditions. Saikosaponin d may display its antitumor effects by influencing the HIF-1α/COX-2 pathway.关键词
