Abstract
Aim To investigate the angiogenesis effects of MOO on HUVECS induced by hypoxia-reoxygenation ( H/R )injury.Methods The models of H/R injury were made with the 3 ~ 8 generation of cultured HUVECS.Cultured HUVECS were divided into six groups: sham group; hypoxia-reoxygenation ( H/R ) group; H/R + SBW; H/R + low dose of MOO( 0.15 g· L-1 ); H/R + medium dose of MOO( 0.15 g · L-1 );H/R + high dose of MOO( 0.45 g· L-1 ).The state of HUVECS was observed with inverted microscope; cell cycle distribution was examined by flow cytometry ( FCM ) ; transwell chamber migration assays were used to calculate the number of migrated cells; matrigel tubule formation assays were employed to assess the area of tube formation.Results Compared with sham group , H/R increased the ratio of G2+ S phase ; but the MOO decreased the Ratio of G2+S phase; the MOO increased the number of cell migration; tube formation in the area of MOO groups was increased.Conclusion MOO can improve the state of cells induced by H/R,inhibit apoptosis and impact the angiogenesis of HUVEC induced by H/R.关键词
