褪黑素对HeLa细胞的生长及NF-κB蛋白表达的影响
The influence of melatonin on NF-κB level in HeLa cells

Abstract
Aim To investigate the anti-proliferative activities of melatonin( MT ) and its influence on NFKB level in HeLa cells.Methods The anti-proliferative activities of MT were evaluated by MTT assay and AO/EB staining, and NF-KB level was observed by Western blot analysis.Results The IC50 on HeLa cells by MT for 72 h was 61.71 mg · L-1.HeLa cells stained with AO/EB were damaged in a time-dependent manner by MT 1 g·L-1 for I d,2 d and 3 d.As the time goes on,more necrocytosis were seen and less apoptosis were observed.It was shown that MT could down-regulate NF-KB level in a time-dependent and a dose-dependent manner after treated with MT 0.2 g ·L-1 for 1 d,2 d and 3 d and with MT 0.008.0.04 and 0.2 g · L-1 for 3 days.Conclusion MT could inhibit the proliferation of HeLa cells with a concomitant down-regulation of NF-KB level,which may be related to its antiproliferative activity.关键词
