摘要

Abstract

Aim To study the cytotoxicity of arsenic trioxide ( As2O3 ) to lymphocytes and the antagonistic and protective effects of N acetylcysteine ( NAC ).Methods Lymphocytes were separated from normal human peripheral blood.The cell survival was examined by Trypan blue dye exclusion assay , apoptosis of lymphocytes was detected by electron microscopy and double staining of FITC-Annexin V/PI, and the cellular reactive oxygen species( ROS )level and Bcl-2 expression were analyzed by flow cytometry( FCM ).Results As2O3 markedly inhibited the growth of human lymphocytes, and induced the cells to display the typical apoptotic changes.During As2O3-induced apoptosis of lymphocytes , the expression of Bcl-2 protein decreased and the production of ROS in the As2O3-treated lymphocytes increased.Compared with As2O3-treatment alone , the combined action of NAC and As2O3 markedly repressed the As203-induced proliferation inhibition and apoptosis of lymphocytes, meanwhile.the production of ROS reduced and the expression of Bcl-2 protein was raised in the lymphocytes.Conclusion As2O3 exerts a significant cytotoxicity to human lymphocytes , and NAC antagonizes the As2O3 -induced cytotoxicity of lymphocytes via elimination of ROS and up-regulation of Bcl-2 expression.
