摘要

Abstract

Objective To investigate enterococci isolated from clinical specimens on the high level of aminoglycoside antibiotic resistance and the detection of aminoglycoside modifying enzyme gene. Methods 112 strains of E.faecalis and 118 strains of E. faecium to the high-level gentamicin(500 μg/ml)and high-level of streptomycin (2 000 μg/ml)minimum inhibitory concentration were detected by agar ditution method. The aac (6')/aph (2"),aph(3')-Ⅲ and ant(6)- Ⅰ genes were amplified by PCR. Finally,their genotype were determined by DNA sequencing. Results 112 strains of E. faecalis and 118 strains of E. faecium HLGR rates were 53. 1% and 73.2%, and HLSR rates were 38. 9% and 59. 8%. The drug resistance of the two species to high-level gentamicin and high-level streptomycin was significantly( P <0. 01 ). Randomly selected HLAR E. faecalis and E. faecium each 48 strains to test aminoglycoside modifying enzymes, including aac ( 6' )/aph (2") gene positive strains were 38 and 45, accounting for 80. 2% and 93.8% ;aph(3')-Ⅲ gene positive strains were 14 and 19,accounting for 29. 2% and 39. 6% ;ant (6)- Ⅰ gene positive strains were 15 and 21 ,accounting for 31.3% and 43.8%. Simultaneously detected two kind and two kind of above strains of aminoglycoside modifying enzyme genes were 40, up to 41.7%. Conclusion The high-level aminoglycoside resistant enterococci have become important nosocomial infection pathogens, and the production of AAC(6')/APH(2")is the most common aminoglycoside resistance mechanisms in Enterococcus.
