摘要

Abstract

Objective :To investigate the role of IRAK1 and IRAK4 in tissue factor expression of THP-1 cells treated with anti-β2GPI/ β2GPI complex .Methods :The TF expression of THP-1 cells was detected by Real-time PCR or TF activity kits after stimulating with antiβ2GPI/β2GPI complex .The IRAK1 ,β-IRAK1 and IRAK4 expression induced by anti-β2GPI/β2GPI complex was determined by Western blot . It was also investigated that IRAK1/4 inhibitor could interrupt the TF expression in THP-1 cells stimulated with anti-β2GPI/β2GPI complex . Results :The TF expression (both mRNA and activity ) of THP-1 cells could be significantly up-regulated with the treatment of anti-β2GPI/ β2GPI complex (100 μg/ml) .The IRAK1 ,p-IRAK1 and IRAK4 expression (protein) in THP-1 cells was significantly increased by stimulation of anti-β2GPI/β2GPI complex (100μg/ml) .IRAK1/4 inhibitor (50 μmol/L ) could decrease the levels of TF and p-IRAK1 in THP-1 cells induced by anti-β2GPI/β2GPI complex .Conclusion :IRAK1 and IRAK4 can be activated and contributed to TF expression of THP-1 cells induced with anti-β2GPI/β2GPI complex .
