Abstract
Objective To investigate the effect of rhHSP70 and human cord blood dendritic cells( DCs ) transfect with RNA of esophageal carcinoma on anti-tumor activity of CTL. Methods The RNA of T. Tn cells were prepared by Trizol reagent. The cord blood monocytes were cultured to produce DCs with rhSCF, rhGM-CSF and IL-4. The DCs were collected and transected with tumor cell RNA. CpG ODN was added in culture medium to enhance the DCs vaccine. The mixed lymphocyte reaction ( MLR ) and cytotoxicity of CTL to T. Tn cells was assayed by MTT colorimetry. Results Compared with pretransfect stage, DCs transfect with RNA of esophageal carcinoma cells showed more MHC-Ⅰ, MHC-Ⅱ, CD54, CD 80 and CD86 ( P ＜ 0.01 ). Compared with immature DCs, mature DCs transfect with tumor RNA enhanced tumor cytolytic activities of T lymphocyte. rhHSP70 could significantly increase the number of molecules on surface of activated T lymphocyte. Conclusion The vaccine of cord blood DCs with adjuvant of rhHSP70 can provide effective and specific way for immunological therapy of esophageal carcinoma.关键词
