小檗碱对高脂-高盐-果糖诱致大鼠高血压性肾损伤的保护作用

Abstract
Objective Investigate the protecting effects of berberine on this model. Methods 50 SD rats were randomly divided into normal group and model group. All rats of the model group were fed by enriched high fat-salt-fructose diet for 8 weeks. On the basis of animal blood pressure, hypertensive rats were randomly divided into model control group ( distilled water) , captopril group ( 25 mg · kg-1 ) , berberine high dose group(300 mg · kg-1) and low dose group( 100 mg · kg-1 ) . These rats were fed by further enriched high fat-salt-fructose diet and treated by intragastric administration of related drugs for 4 weeks. Blood pressure was determined at 0 ,4 , 8 , 12 weekend , and after 4 weeks of drugs treatment , getting urine to determine urine protein, taking blood serum to determine blood fat, blood urea nitrogen and serum creatinine. The contents of Na+ -K+ -ATPase in kidney tissues were determined also. Morphological changes of kidney were observed by light microscope. Results Treated with berberine for 4 weeks, elevated blood pressure and heightened levels of blood fat ( TG ,TC , LDL) in model rats were depressed significantly( P < 0. 01 or P >0. 05) , and lowered HDL-C was elevated significantly ( P < 0. 01) ; the levels of urine protein , blood urea nitrogen and serum creatinine as well as content of Na+ -K+ -ATPase in kidney tissues were lowered significantly (P < 0. 01) ; glomerular diameter and hypertrophia could be improved and degree of hemorrhage became extenuation. Conclusions Berberine can lower hypertension and regulate hyperlipemia,improve renal function and morphological integrity for hypertensive renal impairment model rats fed by enriched high fat-salt- fructose diet.关键词
