Effect of HLA-B on the biological behavior of laryngeal carcinoma Hep2 cell

Abstract
Objective To investigate the effect of HLA-B on the occurrence and development of laryngeal carcinoma.Methods RNA interference technology was used to down-regulate the expression of human leukocyte antigen(HLA) -B in Hep2 cells. Cells transfected with siRNA at most effective interference were used as observed group ,while cells with PBS and nonsense siRNA transfection as control group 1 and 2. Then flow cytometry, MTT assay and cell invasion in vitro were used to detect the biological effect of Hep2 cells induced by HLA-B RNA interference. Results The apoptotic rate of Hep2 cells in observed group at RNA interference 7th day was significantly higher than that in the two control groups ,with S phase cell cycle blocked. The proliferation ability and invasion effect were also increased compared with the two control groupe, all P ＜ 0.05. Conclusion Decrease or loss of expression of HLA-B may improve the occurrence and development of laryngeal carcinoma.关键词
