RP-HPLC法检测大鼠血浆大黄酸含量的经时变化
Ongoing Changes of Plasma Rhein Level in Mouse Tested by RP-HPLC
摘要
Abstract
Objective To observe the ongoing changes of plasma thein level in mouse after orally taken QingReJieDu (清热解毒)decoction.Methods The RP-HPLC analysis was carried out with a Kromasil C18 (250 mm×4.6 mm, 5 μm) column and a mobile phase of methanol-0.1％ phosphoric acid solution (V:V=85:15).The flow rate was 1.0 mL/min and the column temperature was 35 ℃.The detection wave length was set at 254 nm.Results One hour after orally taken QingRejieDu decoction in mouse, the plasma thein level reached a peak value, then decreased steadily and it was metabolized entirely in 12 hours.Conclusion The ongoing changes of the content of thein in mouse plasma can be shown by giving QingRejieDu decoction orally.This RP-HPLC is easy and fast to test the body content of some drugs, hence is suitably used in the study of metabolism features of some herbal medicine.关键词
