Abstract
Objective To investigate the change of 4-1BB ligand (4-1BBL) expression of peripheral blood mononuclear cells in patients with acute atherosclerotic cerebral infarction ( ACI ). Methods Flow cytometry and enzyme-linked immunosorbent assay (ELISA) were used to detocte 4-1BBL expression in peripheral blood mononuclear cells and plasma solube 4-1BBL ( s4-1BBL ) protein level in 20 ACI patients ( ACl ) as compared to nineteen atherosclerosis patients and twenty-three normal controls. Serial studies were performed on some of these ACI patients. Results Compared with the expression of 4-1BBL positive cell rate and mean fluorescence intensity (MFI) on CD14 + monocytes and plasma s4-1BBL levels in normal control group [ ( 4. 6 ± 2.4 ) ％, ( 4. 1 ± 2.7 ), 75 pg/ml] ,the ACI group [ ( 7.9 ± 4. 1 ) ％, ( 7.0 ± 4.0 ), 133.2 pg/ml ] increased significantly, and all the differences were significant (all P ＜ 0. 05 ); while no significant differences were found when comparing to the expression of 4-1BBL positive cell rate [ (6.3 ± 5.0 ) ％ ] and MFI on CD14 + monocytes ( 5.2 ± 2.3 ) and plasma s4-1BBL levels (84 pg/ml ) in atherosclerosis group. Compared with acute stage, the expression of 4-1BBL positive cell rate [ (7.6 ± 5.4 ) ％ ] and MFI on CDl4 + monocytes ( 5.6 ± 2.1 ) and plasma s4-1BBL levels ( 1 1 3.2 pg/ml ) showed a tendency of decrease in the recovery stage of ACI, but there was no significant difference. Conclusion The 4-1BBL expression is upregulation in AC1 patients, as an abnormal immune response that may have been induced during early stages of ACI, may play an important role in the process of atherosclerosis-related ischemic stroke.关键词
