Abstract
Objective To establish a method for determining the serum concentrations of theophylline, Phenobarbital, phenytoin sodium and carbamazepine by RP-HPLC. Methods The analytical column was Promasil C18 column(150 mm ×4. 6 mm, 5 μm).The mobile phase consisted of methanol - water(60: 40) at the flow rate of 0. 7 mL/min. The detection wavelength was set at 205 nm and the column temperature was set at 25 C. Results Theophylline, phenobarbital, phenytoin sodium and carbamazepine were separated well under the chromatographic condition. The linear ranges were 0.30 - 180. 00 μg/mL( r = 0. 999 7) for theophylline, 0.42 - 169.60μg/mL( r = 0. 999 9) for phenobarbital, 0.40 - 170. 00 μg/mL ( r =0. 999 2) for phenytoin sodium and 0. 20 - 140.00 μg/mL( r =0. 999 8) for carbamazepine respectively. The mean relative recovery rates of theophylline, Phenobarbital, phenytoin sodium and carbamazapine were 97.3％, 98.0％, 103. 1％ and 99. 3％ respectively. The intra- day and the inter- day RSD all was less than 6％. Conclusion This method is simple, rapid, accurate without interfering with each other, which is suitable for the blood concentration monitoring of therapeutic drugs.关键词
