大菱鲆亲子鉴定的微卫星多重PCR技术建立及应用
Development of Two Microsatellite Multiplex PCR Systems for Turbot (Scophthalmu smaximus) and Its Application for Family Management

Abstract
We developed two microsatellite multiplex PCR systems using 8 microsatellite loci for turbot (Scophthalmus maximus), which is one important aquaculture species in China. The multiplex PCR microsatellite assay was used to estimate the kinship and the genetic diversity of seven turbot families. Based on the genotypes of either known parent or none of them and offspring, all the absent parental genotypes of the seven families were deduced. According to the unique alleles among the families,each family could be distinguished from the others. Cervus 2.0 analysis showed that the combined exclusion probability was 96. 58% if parents were both unknown and the combined exclusion probability was 99. 71% when only one of the parental genotype was known. The result 70 double-blind individuals showed that the combined exclusion probability and parentage analysis allocation rate was 95. 72% and 92. 86% by Cervus 2. 0 analysis, respectively. The UPGAM dendrogram of the 70 individuals indicated that 95. 71% of the individuals from the same family clusted together. The results showed that microsatellite multiplex PCR is an useful tool for parentage determination and genealogical identification of turbot.关键词
