摘要

Abstract

Aim To investigate the effect of the small molecular weight peptide( KP )on the proliferation and differentiation of mouse MC3T3-E1 cells and the expression of NF-KB p50, p65. Methods The cells growth , alkaline phosphatase ( ALP ) activity ,hydroxyproline content and mineralization of MC3T3El were measured by MTT, PNPP, visible spectrophotometry and alizarin red staining respectively. p65 , p50 protein expression in MC3T3-EI cells were detected by Western blot. Results KP enhanced cell proliferation after MC3T3-E1 cells was treated with KP ( 25. 0,50. 0.100 mg · L-1 )for 24 . 48 h and 72 h. Treated with KP at concentrations of 50. 0 and 100 mg . L-1 for 9,12, 15 d. ALP activity was significantly stimulated;and hydroxyproline contents was enhanced after being treamted 12 .24 d. KP had significant effect on mineralization at concentrations of 50. 0 mg . L-1 and 100 mg · L-1. After 50.0 mg · L-1 and 100 mg · L-1 KP treatment for 3 , 12 , 24 , 30 d , the expressions of p65 and p50 in MC3T3-E1 cells were gradually decreased.Conclusion KP induct MC3T3-EI cells proliferation and differentiation by inhibition of the NF-KB pathway.
