Abstract
Objective To investigate the effect of Ghrelin on monocyte chemoattractant protein-1 ( MCP-1) expression in human brain microvascular endothelial cells ( HBMEC) induced by TNF-α and the effects of p-38 MAPK.Methods After being cultured, different concentration TNF-α were used to treat the HBMEC.In Ghrelin pretreatment group, Ghrelin preprocessing (10 ng/ml) was performed an hour before adding the TNF-α (1.0 ng/ml).The expression of HBMEC MCP-1 mRNA was detected by RT-PCR, and the p-p38 expression was investigated by western blot.Results The expression of MCP-1 mRNA in HBMEC was increased when cells were exposed to different concentration of TNF-α with concentration dependent ( P< 0.05 ) , and the p-p38 expression in cytoplasm increased significantly ( P <0.05) .The expression of MCP-1 mRNA and p-p38 decreased significantly after the Ghrelin preprocessing ( P < 0.05 ) . Conclusion Ghrelin might inhibit the MCP-1 expression in HBMEC via p38 MAPK.关键词
