Abstract

Purpose To investigate the effect of miR-133a on breast cancer cell invasion, migration and proliferating potential, and to explore its possible targets and mechanism. Methods Human breast cancer MCF-7 cells were transfected with Hsa-miR-133a inhibitors hy Lipofectamine to reduce the activity of miR-133a. MCF-7 cells transfected with inhibitor negative control ( inhibitor NC ) were cultured as negative control. Cell proliferation and invasion potential were evaluated by MTS kit and transwell invasion assay. Cell migrating ability of MCF-7 was detected by transwell migration assay and wound-healing assay. Then the possible target genes of miR 133a were forecasted by bioinformatics tools. and the function of these genes was analyzed. Results Cells transfected with miR-133a inhibitors revealed no significant change in growth compared to the inhibitor NC transfectants ( P > 0. 05 ). Remarkable enhancement ofcell migration was ohserved in miR-133a inhibitors transfectant after the scratch ( P < 0. 05 ). Transfection of miR-133a inhibitors intoMCF-7 cells led to a significant increase in cell invasion and migration detected hy transwell invasion and migration assay ( P < 0. 01and P < 0. 05 ). The possible target genes of miR-133a were forecasted hy hioinformatics tools, and part of the target genes played a hiological function of promoting cell invasion, migration. Conclusions MiR-133a may negatively regulate the invasion and migration potential of breast cancer cells and may he a potential candidate target for breast cancer treatment. MiR-133a may play a function of regulation of tumor development through some target genes.
