Abstract
AIM: To study the influence of glucose or insulin at different concentrations on the expression of adiponectin in cardiomyocytes.METHODS : Myocardial cells were isolated from the hearts of neonatal Sprague - Dawley rats and primarily cultured.The cells stimulated with glucose at concentrations of 5.5, 20.25 , 30 , and 50 mmol/L respectively were divided into 5 groups and a hypertonic control group was set up.The cells treated with insulin at concentrations of 0, 10 -10 , 10 -9 , 10 -8 , 10 -7 and 10 -6 mol/L were also divided into 6 groups.The mRNA and protein levels of adiponectin( APN ) were determined by RT - PCR and Western hlotting 48 h after the intervention with glucose or insulin.RESULTS: ( 1 ) The expression of adiponectin decreased with the elevation of glucose concentration at the doses lower than 30 mmol/L( P < 0.05 ).When higher than 30 mmol/L, the expression of adiponectin didn't change significantly.The osmotic pressure did not affect the adiponectin expression in cardiomyocytes.( 2 ) The expression of adiponectin decreased with the elevation of insulin concentration at the doses lower than 10 -7 mmol/L( P < 0.05 ).When higher than 10-7 mmol/L, the expression of adiponectin didn't change significantly.CONCLUSION : The expression of APN decreases with the elevation of glucose or insulin concentrations within a certain range of dosage.关键词
