摘要

Abstract

Considering Lmbr1 is a key candidate gene of chicken polydactyly, the study on search SNPs which was influence the polydactyl. By the methods of PCR-SSCP and direct sequencing, two synonymous mutation (c. 1326A> G, c. 1360C> A) and five genotypes (AABB,aaBB,AABb, AaBb, AaBB) in exon 16 of Lmbr1 gene were detected from Beijing-fatty chicken(4- teod and 5-teod). The statistical analysis showed that the a allele gene frequencies was advantage on c. 1326A>C locus of Lmbrl between double 5-teod and mingle-teod chicken group (0.83, 0.81),A allele gene frequencies was advantage in double 4 -teod chicken(0.53). aa genoptype was preponderance in 5-teod and mingle-teod chicken group (0. 650, 0. 625) ,Aa genotype was dominant position in double 4-teod chicken(0.42), AA was only found on 4-teod chicken group,and was not found in 5-teod and mingle-teod chicken group. B allele gene frequencies was advantage on c. 1326A>C locus in double 5-teod, mingle-teod and double 4-toed chicken group(0.85, 0.88, 0. 76), BB genoptype was preponderance in double 5-teod, mingle-teod and double 4-toed chicken group (0. 692, 0. 750, 0. 516), and was not found bb genoptype. In the chicken group, by Hardy-Weinberg equilibrium tested, c. 1326A>C locus was in non-equilibrium, c. 1360C> A locus was in equilibrium. The research showed that c. 1326A>G was close associate with polydactyly,the site may utilize to MAS.
