Effect of hydroxysafflor yellow A on mitochondrial permeability transition in pneumonocytes after ischemia/reperfusion injury in rats

Abstract
AIM: To investigate the effects of hydroxysafflor yellow A ( HSYA ) on mitochondrial permeability transition in pneumonocyte after ischemia/reperfusion ( I/R ).METHODS: The single lung in situ ischemia/reperfusion animal model was established by opening the chest, blocking the left hilum of the lung for 45 min and unclamping for specified time in rats.Fifty SD rats were randomly divided into 5 groups with 10 rats each: control group, ischemia/reperfusion 1 h group ( I/R 1 h ), I/R 3 h group, and ischemia/reperfusion 1 h + HSYA group ( SI 1 h ) and SI 3 h group.The rats in SI groups were injected with HSYA at the dose of 2.0 mg/kg 20 min before ischemia and at the moment of reperfusion, respectively, while the rats in control group, I/R 1 h group and I/R 3 h group were injected with the same volume of normal saline.Wet to dry weight ratio ( W/D ) of the lungs, the index of quantitative assessment of histological lung injury ( IQA ), apoptotic index ( AI ), the level of cytochrome C ( CytC ) and apoptosis - inducing factor ( AIF ) in endochylema and in mitochondrion, and the function of mitochondrial permeability transition( MPT ) were measured.RESULTS: Compared with control group, significant increases in AI, W/D, IQA, the level of CytC and AIF in endochylema, and significant decreases in the levels of CytC and AIF in mitochondrion were observed in all other groups.AI, W/D and IQA in SI groups were significantly lower than those in I/R groups.Treatment with HSYA markedly reduced the release of CytC and AIF from mitochondrion.There was a significant increase in absorbance at 540 nm in SI 1 h group and SI 3 h group, compared with the corresponding I/R 1 h group and I/R 3 h group.CONCLUSION: By maintaining the function of MPT, decreasing the release of CytC and AIF from mitochondrion to endochylem and preventing the apoptosis of pneumonocytes,HSYA attenuates the injury of lung tissues in the processes of ischemia/reperfusion.关键词
