活菌H37Ra与灭活菌H37Rv感染小鼠腹腔巨噬细胞的实验研究

Abstract
Objective :To study the difference in the expression of immune substances which macrophages are induced by inoculating micewithviable M,tuberculosis H37Ra and Heat-Killed M,tubercubsisH37Rv,and to compare the difference between viable and H eat-K illed M .tubercubsis in the induction of immune responses ,and thus to learn whetherviable or Heat-Killed bacteria could be a possile candidate for new vaccines .Methods :Peritoneal macrophages from BALB /c mouse were cultured with H 37Ra and H 37Rv for 10 hours respectively ,to observe the situation of each group macrophages ,and calculate the phagocytic rates of macrophages by 1he acid dye staining method ,BALB /c m ice were abdominally immunized withviable H37Ra,Heat-Killed H37Rv and NS ,respectively,At30 days after immuniation ,the macrophages were isolated .The expression levels of IL-12P40 /TNF-α ,IFN-γ were detemined by RT—PCR and ELBA .The expression levels of NO ,H202were deternined by Griess and Chemicalmethod ;and 1he changes of CD40L hduced IFN-γ were detected by flow cytometry .Results :The phagocytic rates of macrophages were (55.71±8.42)% for viable M ,tubercubsis and (14 .82±2 .12)% for Heat-Killed M .tubercubsis with a significant difference ( P<0 .01) .Them RNA levels of IL-12P40 ,TNF-α, IFN-γ were higher in viable M . tubercubsis H 37R a than those of in Heat-Killed M .tubercubsis ,1he cytokine levels of IL-12P40,TNF-α,IFN-γ,NO and H2O2were higher in ViableM .tuberculosis H37Ra than those of in Heat-Killed M .tuberculosis .A t the sametime ,1he expressions of CD 40L on macrophages induced by IFN-γ in viable M .tuberculosis stimu-lated group was higher than that of Heat-Killed M .tuberculosis stinulated group .Conclusion:Viable M .tuberculosis H37Ra can induce the activation of macrophages and the production of more protective inmune substances ,which play in portent roles in the host immune response ,so it would be a candidate for tuberculosis vaccine .However,H eat-Killed M .tuberculosis H37Rv can't induce the acttration of macrophages effec-and produce less protective immune substances ,w hich makes it unlikery toi be a candidate for tuberculosis vaccine .关键词
