一种体外扩增γδT细胞的新方法
A novel amplification method of γδT cells
摘要
Abstract
Objective:To establish a novel amplification method of γδT cells.Methods:PBMCs of healthy donors were co-incubated with Zo ledronate-pretreated antologous immature dendritic cells by the ratio of 11∶1 in RPM IL640 completemedium containing 400U/ml IL-2 and 10 ％ newborn bovine serum ,The amplilfication fold of γδT cells were determ ined by MTT assay.The purity and Granzyme B,Perforim and CD 107a expression ampification of γδT cells were verified by flow cytometer.The cytotoxic activity of γδT cells were analyzed by LDH assay.Results:On the 12th day ,the amplification of γδT cells were (120±15 )folds ,on the 8th day and the 11 th day dhe purity of γδT cells reached 70.26％±3.56％ and 90.11％±4.67 resoectively.On the 8th day Granxyme B,Perforin and CD107a expression of γδT cells were 81.66±4.32％、86.62％±5.21％ and 80.12％±3.78 respectively,they were signficantly higher than the control group.The cytotocivity of expanded γδT cells against SGC-7901,SW-1990,SW480 cells reached the peak on the 8th day.Conclusion:γδT cell anpification could be efficiently induced by this method the Granzyme B,Perforin and CD107a expression of γδT cells were significantly higher than the control group,and expanded γδT cells have higher antitum or activity.关键词
