Abstract

OBJECTIVE: To study the effect of total polyphenol of Rhodiola sachalinensis on high fat diet-STZ induced type 2 diabetic rats.METHODS: 36 wistar rats were randomly divided into 6 groups: normal control group (constant volume normal saline), model group (constant volume normal saline), Metformin hydrochloride tablet group (0.3 g· kg-1), R.sachalinensis total polyphenol high-dose, medium-dose and low-dose groups (100,50,25 g ·kg-1).Those groups were given medicine intragastrically once a day for 21 days.12 h after last administration, blood samples were taken from tail vein.The blood glucose (BG) level was determined.Abstaining from food but not drink for 18 h, the activity of SOD and the content of MDA in pancreas cell mitochondria were determined.The anti-hyperglycemic effect of R.sachalinensis total polyphenol was evaluated.RESULTS: BG level of model group was significantly higher than that of normal control group (P<0.01).Compared with model group, BG levels of R.sachalinensis total polyphenol low-dose, medium-dose and high-dose groups decreased significantly (P<0.01), but there was no significant difference between R.sachalinensis total polyphenol middle-dose group and Metformin hydrochloride tablet group.Compared with normal control group, the content of MDA in pancreas cell mitochondria in model group increased significantly (P<0.01 ), but SOD activity significantly reduced (P<0.01).Compared with model group, the content of MDA in R.sachalinensis total polyphenol groups reduced significantly (P<0.01), and SOD activity increased significantly (P<0.01).CONCLUSION: Total polyphenol of R.sachalinensis have good anti-hyperglycemic effect, and it is probably due to antioxidant effect of polyphenols in it.
