异莲心碱在大鼠体内的药动学研究

Abstract
OBJECTIVE: To establish a method for pharmaceutical study of isoliensine in rat. METHODS: RP-HPLC method was used to determine the concentration of isoliensine. The determination was performed on a Hypersil C18(250 mm×4.6 mm, 5 μm) column with mobile phase consisted of methanol-acetonitrile-0.05 mol· L-1 KH2PO4(22:38:40) (pH adjusted to 7.5 with triethylamine) at a flow rate of 1.0 mL· min-1. The detection wavelength was set at 280 nm. RESULTS: The linear equation was A=63.043c+3.937(r=0.999 8). The linear range of isoliensine was 0.50～40.00 μg· mL-1.The lowest detection limit was 0.10 μg· mL-1.The RSD of intra-day and inter-day were below 4.10％. The recoveries of the control samples at low, medium and high concentrations were above 91.20％. CONCLUSION: The method is rapid, simple and accurate, and it is suitable for the pharmacokinetics study of isoliensine in rats.关键词
