Content Determination of Luteolin in Callicarpa kwangtungensis and Callicarpa macrophylla in Different Harvesting Time by RP-HPLC

Abstract
OBJECTIVE: To determine the content of luteolin in Callicarpa kwangtungensis and Callicarpa macrophylla in different harvesting time and to explore the rules of dynamic change of luteolin content. METHODS: RP-HPLC was performed on Alltech C18(250 mm×4.6 mm, 5 μm) column with column temperature at 30 ℃. The mobile phase consisted of methanol-0.3 % phosphate aqueous solution (47: 53) at flow rate of 1 mL·min-1, and the detection wavelength was set at 352 nm. RESULTS: The linear range of luteolin was 0.015~0.300 μg (r=0.999 9) with an average recovery of 98.52% (RSD= 1.84% ,n=6). The content of luteolin increased gradually with the growth of plants and reached the peak at the end of growth period. The content of luteolin in C.macrophylla was commonly higher than in C. kuangtungensi. CONCLUSION: Results of study can provide scientific basis for natural cultivation or artificial cultivation of C. kwangtungensi and C. macrophylla.关键词
