摘要

Abstract

[ Objective ] The aim to study the gus gene transformation transient expression on microneedle injection sweet corn germ, to lay basis for deeply researching corn transformation. [ Method ] The experiment was done by using gus gene transient expression technology mediated by agrobacterium, and a embryo injection system for open gene transformation was built. [ Result ] The results showed that the seeds germ with the length of 3 mm was the most optimal transformation receptor. The optimal OD value of EHA105 agrobacterium of bacterial concentration was 0.558 -0.630. The optimal days of co-culture in gus gene transformation was 3 days. Under the optimal conditions, the microneedle injection of germ growth point gene conversion rate was 1%. [ Conclusion] The study can provide reference for improving the hereditary character and cultivating new varieties of genetically engineering of sweet corn.
