东方百合高效离体再生体系的建立
Establishment of an Efficient Micropropagation System for Oriental Hybrid Lily
摘要
Abstract
A two-step explant method combined with embryogenic callus formation which could greatly increase regeneration efficiency of Oriental Hybrid lily was developed. Bulb scale segments were used as original explants and shoot sections excised from shoot clusters were used as the secondary expLants.ln addition,the effect of list range of 2,4-D(2,4-Dichlorophenoxyacetic acid) concentration and the effect of different kinds of secondary explants on calli induction have been studied. Moreover, the critical concentration of G418 has been determined.Our results indicated that the propagation efficiency could be as high as 27 000 times. The whole progress of from bulb scale to new plant formation only need 9 months. The medium supplement with 2.0 mg/L 2,4-D in conjunction with short basal stem segment as secondary explant was most favorable for embryogenic calli formation and the critical concentration of G418 was 50 mg/L.关键词
