Abstract
Objective To investigate the mechanisims of complement C3 during the acute phase of intracerebral hemorrhage ( CH ) in rats and its role in the increasing of brain water content; to investigate the effects of cobra venom factor ( CVF )on C3mRNA expression and brain water content after intracerebral hemorrhage in rats. Methods Rat CH model was established by injecting autologous blood into the caudate nucleus of rats stereotaxically. SD rats were divided into three groups ( hemorrhagic group, sham operation group and CVF treatment group ) and sacrificed 3 h, 6 h, 12 h, 1 d, 3 d, 5 d, and 7 d after establishment of the model. Brain water content ( BWC ) was determined using wet and dry weight measurement of and mRNA levels of C3 were determined by RT - PCR method. Results Expression of C3mRNA increased 6 hours after CH and peaked 3 days after CH.Brain water content around the hematoma increased 3 h after CH and peaked 3 days after CH ( P < 0.05 ), which decreased to normal levels at 7 d time. Positive correlations existed hetween brain water content and C3mRNA expression ( r = 0.758, P <0.05 ). C3mRNA expression around hematoma decreased significantly after CVF intervention in comparison with the hemorrhage group ( P < 0.05 ). Brain water content in the intervention group was significantly lower than in the CH group ( P < 0.05 ). Conclusion The complement cascade may be activated after CH with the expression of C3mRNA increased significantly, CVF intervention, which reduces C3mRNA expression and brain edema, exhibits a protection effect on the damaged brain tissue after CH.关键词
