阿魏酸对人角质形成细胞的保护作用
Protective Effects of Ferulic Acid on Human Keratinocyte
摘要
Abstract
Objective To investigate the protective effects and mechanism of ferulic acid on human keratinocytes( HKC ). Methods The cell model of oxidative stress was established in HKC treated with CoCl2. The cell viability post cultured with ferulic acid was detected by MTT method, the secretion of cytokines TNF-α and IL-8 in the supernatant was measured by ELISA. Results The surviving rate of HKC cell with normal group was 100.00% and the contents of TNF-α and IL-8 was 11.37, 98.30 pg · mL-1 respectively. The surviving rate of HKC cell respectively was 96.15% ,67.92% in group treated with 0.03 and 0.3 mg · mL-1 Ferulic acid,which were significantly higher than those in the model group (46.20%, P<0.01 ,P<0. 05 ), and the effect was in a dose-dependent manner. The contents of TNF-α( respectively were 12.95 ,17.46 pg · mL-1 )were significantly lower than those in the model group ( 29.54 pg · mL -1 , P < 0. 01, P < 0. 05 ), The contents of IL-8 ( respectively were 102.76, 128.96 pg · mL-1 )were significantly lower than those in the model group( 162.33 pg · mL-1 ,P<0. 01 ,P<O. 05 ). However, There were no significant effects on the surviving rate of HKC cell and the contents of TNF-α and IL-8 in group treated with 0.003 mg· mL-1 Ferulic acid. Conclusion Ferulic acid may prevent HKC against oxidative injury by inhibiting secretion of TNF-α and IL-8.关键词
