Packaging and identification of lentivirus granules interfering GATA2
摘要
Abstract
Objective To prepare the lentivirus granules interfering GATA2 and establish the cell line K562 which downregulate GATA2 expression consistently. Methods The DNA segments carried the specific GATA2 interfering sequence into the plasmid PsicoR. The recombinant plasmid was transfected into the K562 cell and the interfering effect was investigated by Western blot. The siGATA2 plasmid and three packing plasmids plPI , plP2 , pl/VSVG were transfected into HEK293T cell to produce lentivirus granules interfering GATA2. The siGATA2-K562 cell line was established after infecting the lentivirus granules interfering GATA2 and the interfering effect was investigated by Western blot. Results The plasmid and lentivirus granules interfering bearing the specific interfering sequences was successfully prepared, and the expression of endogenous CATA2 was down-regulated by the siGATA2 lentivirus. Conclusion The lentivirus granules interfering GATA2 are successful prepared. The stable siGATA2-K562 cell line is established.关键词
