Effect of intrathecal injection of nerve growth factor on functional recovery and nerve cells after spinal cord injury in rats
摘要
Abstract
Objective To study the effect of intrathecal injection of nerve growth factor( NGF ) on functional recovery and nerve cells after spinal cord injury( SCI ). Methods 52 rats were randomly divided into three groups: intrathecal injection of NGF group( n = 20 ), control group( n = 16 ) and musclar injection of NGF group( n = 16 ).After SCI was made in all these rats at T10 with modified Allen’s method , the same volume of NGF solution ( about 1. 5 μg pure NGF ) and NS were intrathecal injection into the damaged areas respectively by the equipment which could afford a minimal injection. In the I ,7 ,14 ,21 ,28 day after SCI. BBB rating scale was used to observe the recovery of motor function. In the 7,14,21 ,28 day after SCI, immunofluorescence and histopathologic examinations were performed to study the number of the adult endogenous neural cells and pathological changes. Results From 24 h t0 4 weeks after SCI. the BBB scores of intrathecal injection of NCF group was higher than that in control group and musclar injection of NGF group. From 21 day, the scores of the intrathecal injection of NGF group was significantly higher than that in control group. Histologic observation showed that the morphology of spinal cord and neurons in intrathecal injection of NGF group was better than that in control group and musclar injection of NGF group. The mean number of endogenous neural stem cells( NSCs ) was more than that in control group and musclar injection of NGF group,while astrocytes was less than that in the other two groups. Conclusion Intrathecal injection of NCF could significantly increase the number of endogenous NSCs and promote them differentiate into neurons. thereby could stimulate effectively nerve occurrence and motor functional recovery after SCI.关键词
