Abstract
Purpose The purpose of this study was to detect COX-2 , iNOS expression in colorectal carcinoma( CRC ). This may provide theories for the research of inflammation and the colorectal carcinomas. Methods The expressions of COX-2 , iNOS were studied in 100 cases of human colorectal carcinomas and 11 cases of normal colorectal mucosa by flow cytometry ( FCM ) and immunohistochemical staining. The clinical pathological significance of COX-2 , iNOS expression in CRC were also analyzed. Results Immunohistochemistry showed that the positive rate of COX-2 expression in colorectal carcinoma was 76. 00% while in normal mucosa was only 9. 10% , the expression of COX-2 in two groups were significantly different ( P < 0. 05 ). The positive rates in lymph node metastasis and without lymph node metastasis groups were 87. 50% ( 42/48 ), 65. 38% ( 34/52 ) respectively. There was significant difference between the two groups too( P < 0. 05 ). The mean FI values of COX-2 expression detected by FCM in well, moderately and poorly differentiated groups were 2. 47 ± 1. 41 , 2. 70 ± 1. 08 and 3. 01 ± 1. 26 , respectively. The expression in each group was significantly higher than normal mucosa ( 1. 00 ±0. 28 )( P < 0. 01 ). Also, COX-2 expression in lymph node metastasis group ( 2. 45 ± 1. 41 ) was significantly higher than the group without lymph node metastasis ( 1. 61 ± 1. 27 ) ( P < 0. 05 ). The positive rate of iNOS expression in colorectal carcinomas ( 81. 00% ) was significantly higher than in the normal mucosa ( 27. 27% )( P < 0. 05 ), and positive rate in lymph node metastasis group ( 91. 67% ) was significantly higher than the group without metastasis ( 71. 15% )( P < 0. 05 ). Closely correlation was found between COX-2 and iNOS expression ( P < 0. 05 ). Conclusions Expressions of COX-2 , iNOS in colorectal carcinomas were abnormally increased and were closely related with lymph node metastasis. It may play an important role in the progression and metastasis of colorectal carcinoma.关键词
