Abstract
Objective To explore the effect of sodium fluoride on apoptosis and expression of caspase-3 of mouse Leydig cells cultured in vitro. Methods Leydig cells cultured in vitro were divided into 4 groups, and then were treated with 0.5 ,10,20 mg/L of sodium fluoride , respectively. Annexin V -PI double staining with flow cytometry ( FCM ) was applied to detect Leydig cell apoptosis in each group after treatment for 48 h. The expression of caspase-3 was detected hy immunohistochemistry. Results Apoptosis and caspase-3 expression were found after treated with sodium fluoride. Apoptosis rate was significantly higher in 20 mg/L sodium fluoride group than in the other groups, and the gray value of caspase-3 expression was significantly lower( P ＜0. 01 ). Conclusion The results suggest that fluoride could promote Leydig cell apoptosis and caspase-3 expression in vitro , and have cvtotoxic effect on mouse Leydig cells.关键词
