Abstract
[ Objective]The aim was to study the mechanism of inflammatory response caused by NH4 +. [ Method ] By using inverted microscope,the variation of monocyte THP-1 morphology caused by different ionic salts were detected,moreover,based on LDH kit to detect LDH in cell culture supernatant,what's more, through trypan blue staining to detect cell morphology and survival, and the production and secretion of inflammatory cytokine were detected by Western blot. [ Result]In vitro study,we found that NH4 + could activate caspase-1 and downstream cytokine IL-1β,IL-1β was an important inflammatory cytokine;K+ can inhibit caspase-1 and IL-1β activation. [Conclusion]Na+ ,SO2-4 ,CI- irons did not cause inflammation of the THP-1 cell;NH4 + can cause monocyte cells inflammation,and the inflammation can be inhibited by K+ ion,which provided certain basis for disease treatment caused by amino acid metabolism disorders.关键词
