The Effects of Ganoderma Lucidum Spore Oil on MCF -7 Cell Apoptosis and Migration

Abstract
Objective: To observe the effects of Ganoderma lucidum spore oil on proliferation, apoptosis and migration of human breast cancer cell MCF - 7 in vitro, and explore its possible mechanisms. Methods: The prolfferation inhibition rate of MCF - 7 cells was determined by using the cell count kit 8 ( CCK - 8 ). Wound healing assay was used to evaluate the cell migration. Cell apoptosis was detected by flow cytometry. Furthermore, to analyze the expression level of Bcl - 2, Bax and VEGF mRNA, RT - PCR was performed. Results:Different concentrations of Ganoderma lucidum spore oil could inhibit MCF -7 cell proliferation as well as migration and took on dose and time dependently. It also induced cell apoptosis in a dose dependent way . The mRNA expressions of Bax related to apoptosis were increased, while Bcl -2 was decreased. On the other hand, the VEGF mRNA expression, which was important to tumor metastasis, was also declined. Conclusion:Ganoderma lucidum spore oil can inhibit MCF -7 cell proliferation and migration, on the other hand, it induces cell apoptosis. The mechanisms may be partly due to the decreased Bcl -2/Bax rate and VEGF mRNA expression.关键词
