甘露醇对骨髓间充质干细胞治疗血管性痴呆大鼠行为学的影响
THE USE OF MANNITOL FOR BONE MARROW MESENCHYMAL STEM CELLS TRANSPLANTATION AND ITS EFFECT ON BEHAVIOR IN VASCULAR DEMENTIA RAT MODEL

Abstract
Objective:To investigate the use of mannitol intravenous injection for bone marrow mesenchymal stem cells(BMSCs) transplantation and its effect on changes of behavior in vascular dementia rat model. Methods:Rats of experimental group were treated with a permanent bilateral common carotid arteries (CCA) occlusion (2-VO) to establish vascular dementia model. After the modeling, experimental animals were divided into culture media group(intravenously injecting serum-free growth medium 1 mL), BMSCs transfer group(intravenously injecting 1 million BMSCs in 1 mL growth medium) and mannitol+BMSCs transfer group(intravenous injection of mannitol 1.5 g/kg in PBS followed by intra-arterial injection of 1 million BMSCs in 1 mL growth medium). The changes of rats' behaviors were detected by Morris water maze in four groups 4 weeks after transplantation. Results:Comparing to culture media group, the escape latency decreased significantly( P <0. 05) and platform quadrant residence time increased significantly( P <0.05) in BMSCs transfer group and mannitol + BMSCs transfer group. Comparing to BMSCs transfer group, the escape latency decreased significantly (P <0.05) and platform quadrant residence time increased significantly ( P <0. 05) in mannitol+BMSCs transfer group. Conclusion:The combination of mannitol and BMSCs transplantation significantly increased learning and memory function in vascular dementia in rats, and the mechanism associated with mannital's liberalizing BBB and facilitating the migration of BMSCs into ischemic brain tissue and its survival.关键词
