芥菜籽提取物对人结肠癌细胞系SW480凋亡的影响
Influence of mustard seed extract on human colorectal cancer cell line SW480 apoptosis
摘要
Abstract
Objective To observe the effects of mustard seed extract (MSE) on human colorectal cancer SW480 cellular apoptosis, primarily explore its possible mechanism. Methods Take cell culture in vitro. The effects of MSE on cell proliferation were observed by CCK-8 method in vitro. The morphologic changes of apoptosis cell were evaluated by Hoechest33258 fluorescent staining. The cell apoptosis was tested by flow cytometer, and the activity of Caspase-3 was tested by Caspase-3 fluorometric assay kit. Results MSE exhibited a marked anti-proliferative effect on SW480 cells within the concentration from 0.2 to 1.0 mg/ml and the time from 24 to 72 hours( P ＜0.01 ). The apoptotic ratio was dose-dependant in 24 hours. The activity of Caspase-3 had a significant increase compared to the control group in 24 hours ( P ＜ 0.05 ).Conclusions MSE can inhibit the proliferation of SW480 cell effectively. The effect may be correlated with the tumor cell apoptosis and the Caspase-3 activity induced by MSE关键词
