Abstract

OBJECTIVE: To prepare chloramphenicol thermosensitive in-situ gels for ophthalmic use and to evaluate erosion and eye irritation character of gel in rabbit. METHODS: The thermosensitive gels were prepared by cold-solving method, and the content of chloramphenicol was determined by HPLC. The drug release of the formulation was investigated with a membrane-less dissolution model and drug release mechanism was explored. Eye irritation experiments were performed on rabbit by eye instillation for 7 days. RESULTS: Prepared gels were liquid at room temperature and turned into semi-solid gels at body temperature. The in vitro release experiment showed that the release of chloramphenicol from poloxamer gels followed Hixon-Crowell kinetics while the erodible rates of gel accorded with zero-order model. The drug release mechanism included drug diffusion and gel erosion. The irritation score of gels was lower than 3 after administration. R suggested that the thermosensitive in-situ gel containing chloramphenicol was nonirritant. CONCLUSION: Prepared formulation meets the demands of thermosensitive in-situ gel for ophthalmic use.
