摘要

Abstract

Aim To study the effects and mechanismsof tangeretin combined with 5-fuorouracil ( 5-Fu ) onproliferation of human AGS gastic cancer cells. Meth-ods MTT assay was used to detect the growth inhibi-tory rates of human AGS gastric cancer cells treatedwith 5-Fu and tangeretin combined with 5-Fu. The cellcycle distribution was measured by flow cytometry. Theprotein expressions of p21 and p53 were detected byWestern blot. Results MTT assay showed 5-Fu inhib-ited the growth of AGS gastric cancer cells in a dose-dependent manner ( P < 0. 05 , P < 0. 01 ) and inhibi-tory rates of tangeretin combined with 5-Fu was higherthan those of 5-Fu alone ( P < 0. 01 ). Tangeretin com-bined with 5-Fu arrested AGS cells in S phase com-pared to control group ( P < 0. 01 ). Western blotshowed that tangeretin combined with 5-Fu increasedexpressions of p53 and p21 protein compared to thattreated with 5-Fu alone ( P < 0. 01 ). ConclusionsTangeretin combined with 5-Fu can enhance the prolif-eration inhibition of 5-Fu in human gastric cancer AGScells. S phase arrest and upregulation of p53 and p21protein might be the mechnisms.
