Effects of elemene on proliferation and topoisomerase of human hepatocarcinoma HepG-2 cells
摘要
Abstract
Aim To investigate the effects of elemene ( EIE ) on the proliferation, apoptosis and expression of topoisomerase Ⅰ and TOPO Ⅱ( TOPO Ⅰ and TOPO Ⅱ ) of hepatocarcinoma HepG-2 cells. Methods After treatment with ELE , hepatocarcinoma HepG-2 cells was observed under inverted microscope. Cell proliferation was assessed by MTT assay. Apoptosis was detected by Annexin V/PI staining. TOPO Ⅰ and TOPO Ⅱ mRNA were analyzed by RT-PCR. Expressions of TOPO Ⅰ and TOPO Ⅱ protein were analyzed by WesternBlot. Results ELE significantly inhibited HepG-2 cell proliferation in a dose- and time-dependent manner. induced apoptosis and down-regulated mRNA and protein expressions of TOPO Ⅰ and TOPO Ⅱ in a dose-dependent manner. Conclusion ELE can inhibit proliferation of human hepatocellular carcinoma HepG-2 cells. induce tumor cells apoptosis. The down-regulation of TOPO Ⅰ and TOPo Ⅱ expressions might be one of the mechanisms of ELE anti-tumor.关键词
