摘要
Abstract
To investigate the effect of deoxynivalenol (DON)on the mitochondrial membrane potentials MMP) and Bax, Bcl-2 expression of hamster kidney cells (BHK-21 cell )in vitro, the change of early apoptosis, mitochondrial membrane potential and apoptosis-related proteins with Bax and Bcl-2 expression were measured by flow cytometry and immunocytochemical staining assay. The results showed that different concentration of DON could induce apoptosis for BHK-21 cells within the experimental concentration and cause apoptosis significantly as compared with control group (P < O. 05 ). A marked decrease of mitochondrial membrane potential in a dose and time dependent manner were observed clearly by mitochondrial membrane potentials assay. Immunohistochemical staining analysis showed that, after 24 h treatment with DON, the expression of Bax was significantly higher, while Bcl-2 decreased. These result suggested that DON could induce apoptesis and the MMP disruptionin of BHK-21 cells in vitro. The essential mechanisms of apoptosis might be related to inducing the change of hepatocarcinoma cell cycle, the expression of the Bax increase and Bcl-2 decrease in BHK-21 cells.关键词
