Abstract

To construct a stable genetic transformation system, the protoplasts of Mucor circinelloides sp.EIM- 10 spores were prepared by using enzymatic methods. The influences of enzyme system, osmotic pressure stabilizer, enzymolysis time, and temperature on the preparation and regeneration of the protoplasts of Mucor circinelloides sp. EIM-10 spores were studied. The optimum preparation conditions were established by using enzyme system composed of 1% cellulase and 2% lywallzyme with 0. 5 mol/L NaCl being used as osmotic pressure stabilizer at 32 ℃ for 2. 5 h. When regenerated,0. 5 mol/L NaCl was chosen as osmotic pressure stabilizer by using the double-decked culture medium. Under the conditions, the formation of protoplast was 1.2 × 106/mL and the regeneration rate was 70.5%.
