Abstract

OBJECTIVE: To develop the method for concentration determination of biapenem in human plasma and study its pharmacokinetics.METHODS: Plasma samples were processed by 7% zinc sulfate solution, and HPLC method was adopted.The determination was performed on Agilent Zorbax Bonus-RP(250 mm×4.6 mm, 5 μm)column with mobile phase consisted of methanol-0.03% glacial acetic acid (gradient elution) at a flow rate of 1.0 mL·min-1.The detection wavelength was set at 300 nm.The pharmacokinetic parameters were calculated by DAS 2.0 software after single dose and multiple dose of biapenem.RESULTS: The linear range of biapenem was 0.2~50 μg·mL-1(r=0.999 8) with an average recovery of 96.8%~99.4%.The RSD of intra-day and inter-day were all lower than 8 %.Main pharmacokinetic parameters of single intravenous dose of biapenem with low, medium and high dose were as follows: t1/2( 1.47 ± 0.23)h, ( 1.38 ± 0.28)h, ( 1.52 ± 0.06)h; cmax(8.92 ± 4.13)mg· L-1, ( 19.07 ± 3.08) mg· L-1,(35.60 ± 0.05)mg·L-1; AUC0~t(11.17 ± 3.01) mg·h·L-1, (22.14 ± 3.97) mg·h·L-1, (45.26 ± 3.85) mg·h·L-1.The pharmacokinetic parameters of multiple-dose of biapenem were as follows: t1/2 ( 1.59 ± 0.13) h, cmax( 18.96 ± 6.03)mg· L-1, AUCss(24.5 7 ± 2.81 ) mg·h·L-1.CONCLUSION: The good linearity of single intravenous dose of biapenem is obtained in the range of 150~600 mg.There is no accumulation of biapenem in plasma after multiple dose of biapenem.
