柱前衍生化HPLC-UV法测定天麻胶囊中齐墩果酸的含量
A pre-column derivatization HPLC-UV for determining oleanolic acid in Tianma capsule
摘要
Abstract
Objective To develop a pre-column derivatization HPLC method for determining the contents of oleanolic acid in Tianma capsule. Methods The oleanolic acid was benzoylated by paranitrobenzoyl chloride as derivatizing reagent, and derivatives of oleanolic acid were determined by HPLC-UV. A Diamonsil C18 column ( 200 mm × 4. 6 mm,5 μm )was used with the mobile phase of acetonitrile -0.2％ phosphoric acid( 95∶ 5 ) at the detection wavelength of 254 nm. Results The linear regression equation of derivatives of oleanolic acid was Y = 90 374X +90. 208( r = 0. 999 ). The content of derivatives of oleanolic acid showed a good linearity within the range of 0. 39 - 6. 20μg. Conclusion The established pre-column derivatization HPLC-UV method can be used for determining the content of oleanolic acid. The derivative is p-nitrobenzoic acid ester-olean. The UV wavelength should he red-shifted to 254 nm. The method is accurate and sensitive.which offer a scientific basis for controlling the quality of Tianma capsule.关键词
